The Amnesia Plot

Many books (such as The Bourne Identity), movies (such as Dark City), and certain TV show episodes (including ones in Buffy and Xena) use character amnesia as a starting point. If done well, the writer can make a salient point about what is truly important about self-definition. If done poorly, the writer can lose the audience due to lack of a connection with the character. How does a writer take this gimmick and turn it into an awesome experience for the reader/audience?
